Which way do I slice brisket?

Across the grain, and the grain runs differently in the point and the flat. Separate the two, turn the point ninety degrees, and slice each across its own grain.

How thick should brisket slices be?

About the thickness of a pencil for the flat, and cubes or thicker slices for the point. Slices should hold together when lifted but pull apart easily.

Does knife sharpness matter that much?

Enormously for slicing. A dull blade compresses and tears the meat, releasing juice onto the board rather than keeping it in the slice.
